Hematoxylin and Eosin Staining Kit is the most common staining procedure used for light microscopy. A paraffin or frozen section stuck on a glass slide is rinsed with organic chemicals to dissolve the paraffin or O.C.T compound and leave the sectioned tissue on the glass slide. The tissue is then rehydrated and dipped into an aqueous solution of hematoxylin. Hematoxylin (which is bound to carrier molecules) binds to anionic groups such as the phosphate groups of nucleic acids. Following dehydration in alcohol, the tissue is dipped into an alcohol solution of eosin. Eosin carries a negative charge and reacts with the cationic groups common to amino acids. Once stained, the tissue is covered with a thin glass cover slip attached with a mounting medium. In general, the eosin imparts a pink to red color to proteins, and hematoxylin stains nucleic acids from blue to purple.

Hematoxylin staining solutions usually have long shelf lives. Hematein, the active component of “hematoxylin” stains, stains metal ions; it stains any tissue component to which the mordant metal binds. Hematoxylin and Eosin Staining Kit is optimized for use on cryostat and paraffin-embedded section. This kit is made in a ready-to-use format that eliminates the need for extra steps in the procedure, minimizes hands-on time, maximizes convenience and reduces the risk of stained cell numbers.
