TEMPase Hot Start DNA Polymerase

TEMPase Hot Start DNA Polymerase is useful for performing automated hot start amplification due to thermal activation at a temperature well above optimal annealing.
The enzymes remain inactive until the time and temperature is optimal. During hot start, primers bind only to their specific target, and polymerase activity is directed exclusively to that target.
As a result only the region of interest is amplified, which increases sensitivity and yield while reducing non-specific background amplification.
The result is higher specificity and greater yields when compared to standard DNA polymerases.

Buffer systems for TEMPase Hot Start DNA Polymerase

10X TEMPase Buffer I; -A standard Ammonium Buffer with a final MgCl2 concentration of 1.5mM suitable for most applications.

10X TEMPase Buffer II: -Is Ampliqon’s new recommendation for use with TEMPase Hot Start DNA Polymerase. This is a optimized buffer system with a balanced Ammonium/Potassium concentration with a final MgCl2 concentration of 1.5mM suitable for most applications. This buffer improves more complicated PCR systems such as multiplex PCR.

10X TEMPase Buffer II Tween Free; -This is a optimized buffer system with a balanced Ammonium/Potassium concentration The buffer do not contain tween for those applications, where the products are analyzed on a system, which do not tolerate tween, such as DHPLC (denaturing High-Performance Liquid Chromatography)

Our buffers are also available without MgCl₂ for customers who prefer a Mg concentration below 1.5mM –or just prefer to add MgCl₂ themselves.
